摘要 [目的]確定黃芪與大豆共發(fā)酵制備一種具有高纖溶活性黃芪豆豉的最優(yōu)發(fā)酵工藝。[方法]以黃芪水提液浸泡大豆作為發(fā)酵基質(zhì),以淡豆豉來(lái)源的4株枯草芽孢桿菌作為發(fā)酵菌株,以纖溶酶活性為考察指標(biāo),通過(guò)對(duì)發(fā)酵菌株的篩選、黃芪提取液的濃度和比例以及發(fā)酵時(shí)間的考察,篩選最優(yōu)發(fā)酵工藝。[結(jié)果]黃芪豆豉的最優(yōu)發(fā)酵工藝:黃芪水提液濃度為75 mg/mL,黃芪水提液與大豆的比例為1∶1(mL∶g),最優(yōu)發(fā)酵菌株為DCT-1,于37 ℃發(fā)酵36 h。[結(jié)論]黃芪與大豆共發(fā)酵制備黃芪豆豉,既提高了豆豉的纖溶酶活性,又兼具黃芪的補(bǔ)氣功效,提供了一種具有益氣活血作用的食療產(chǎn)品。

Abstract [Objective]Astragalus membranaceus and soybean were cofermented to prepare fermented soya beans with high fibrinolytic activity, and confirming the optimal fermentation process.[Method]Soybean was soaked in astragalus aqueous extract as the fermentation substrate, and 4 strains of Bacillus subtilis separated from Sojae Semen Praepartum were used as the fermentation strains, with fibrinolytic activity as the index, and the optimal fermentation technology was selected through the screening of strains, the concentration and proportion of astragalus extract and the fermentation time.[Result]The results showed that the optimal fermentation process of Astragalus membranaceusfermented soya beans was as follows: astragalus water extract concentration was 75 mg/mL, the proportion of astragalus water extract and soybean was 1∶1(mL∶g), DCT1 was the optimal fermentation strain, and fermented at 37 ℃ for 36 h.[Conculsion]The cofermentation of astragalus and soybean not only improved the fibrinolytic activity of fermented soya beans, but also had the effect of supplementing breath of astragalus, providing a kind of food therapy product with the function of Yiqi Huoxue.

豆豉為我國(guó)傳統(tǒng)大豆發(fā)酵調(diào)味品,具有一定的藥用價(jià)值[1]。我國(guó)不同地域已形成了很多獨(dú)具特色的豆豉產(chǎn)品,其中潼川豆豉和永川豆豉已被列入國(guó)家級(jí)非物質(zhì)文化遺產(chǎn)[2]。傳統(tǒng)豆豉是在自然環(huán)境條件下由多菌種混合發(fā)酵而成,其風(fēng)味和生理活性物質(zhì)的含量受加入的輔料、生產(chǎn)環(huán)境、生產(chǎn)工藝等因素的影響,如中藥淡豆豉是大豆與青蒿、桑葉或紫蘇、麻黃發(fā)酵后制成因而具有不同的藥性[3-4]。豆豉在自然發(fā)酵過(guò)程中存在污染有害菌的風(fēng)險(xiǎn)[5-6],采用純種發(fā)酵可有效避免此現(xiàn)象發(fā)生。豆豉含有多種營(yíng)養(yǎng)和生理活性成分,其中豆豉纖溶酶可有效溶解血栓,長(zhǎng)期食用可預(yù)防心腦血管疾病的發(fā)生,且無(wú)毒副作用[7],而枯草芽孢桿菌為產(chǎn)豆豉纖溶酶的主要發(fā)酵菌[8]。心腦血管疾病患者常伴有氣虛血瘀癥狀,在改善血液循環(huán)的同時(shí)應(yīng)加強(qiáng)補(bǔ)氣[9]。黃芪為常用的補(bǔ)氣中藥,黃芪多糖可促進(jìn)益生菌芽孢桿菌的增長(zhǎng),芽孢桿菌發(fā)酵黃芪可提高活性成分的含量,增強(qiáng)藥效[10-12]。
筆者以大豆輔以黃芪作為發(fā)酵基質(zhì),采用枯草芽孢桿菌純菌種發(fā)酵制備一種具有益氣活血作用的黃芪豆豉,通過(guò)對(duì)發(fā)酵菌株、黃芪提取液的濃度及比例、發(fā)酵時(shí)間等因素的考察,以纖溶酶活性為指標(biāo),篩選最優(yōu)發(fā)酵工藝,為黃芪豆豉作為食療產(chǎn)品的應(yīng)用提供依據(jù)。
1 材料與方法
1.1 材料
1.1.1 原材料。 黃芪飲片由亳州市張仲景中藥飲片有限責(zé)任公司生產(chǎn),經(jīng)黑龍江省中醫(yī)藥科學(xué)院王偉明研究員鑒定為豆科植物蒙古黃芪[Astragalus membrannceus(Fisch.)Bge.var.mongholicus(Bge.)Hsiao]的干燥根;黃豆由黑龍江省農(nóng)業(yè)科學(xué)院提供,2017年種植采收。
1.1.2 試劑。尿激酶(140604-201224)、牛血清纖維蛋白原(140607-201841)、凝血酶原(140605-201526)均購(gòu)自中國(guó)食品藥品檢定研究院,瓊脂糖購(gòu)自BIOWEST公司。營(yíng)養(yǎng)瓊脂培養(yǎng)基、營(yíng)養(yǎng)肉湯培養(yǎng)基均購(gòu)自青島海博生物技術(shù)有限公司,其他試劑均為分析純。
