新鮮洋蔥汁對(duì)帕羅西汀所致雄性大鼠性功能障礙的影響及其機(jī)制研究

目的 探討新鮮洋蔥汁對(duì)正常雄性大鼠和帕羅西汀所致性功能障礙雄性大鼠性功能和血清睪酮水平的影響,為后期臨床研究提供實(shí)驗(yàn)依據(jù)。方法 將有性行為能力的70只雄性大鼠隨機(jī)分為7組,每組10只。對(duì)照組給予蒸餾水灌胃,0.5 mL洋蔥汁組、1.0 mL洋蔥汁組血清睪酮水平2.0 mL洋蔥汁組分別給予相應(yīng)劑量的新鮮洋蔥汁灌胃,帕羅西汀組給予帕羅西汀懸浮液10 mg/kg灌胃,帕羅西汀加1.0 mL洋蔥汁組、帕羅西汀加2.0 mL洋蔥汁組分別給予帕羅西汀懸浮液10 mg/kg和相應(yīng)劑量的新鮮洋蔥汁灌胃,均1次/d,連續(xù)灌胃20 d。在末次灌胃結(jié)束后3 h,記錄各組大鼠性行為參數(shù)及血清睪酮水平。結(jié)果 1.0 mL洋蔥汁組和2.0 mL洋蔥汁組大鼠的騎跨潛伏期、騎跨次數(shù)均明顯低于對(duì)照組和0.5 mL洋蔥汁組(P均<0.05),交配有效率和血清睪酮水平均明顯高于對(duì)照組和0.5 mL洋蔥汁組(P均<0.05);1.0 mL洋蔥汁組和2.0 mL洋蔥汁組大鼠的插入潛伏期和1.0 mL洋蔥汁組大鼠的射精潛伏期均明顯低于對(duì)照組(P均<0.05);1.0 mL洋蔥汁組大鼠的插入潛伏期和射精潛伏期均明顯低于0.5mL洋蔥汁組(P均<0.05)。帕羅西汀組大鼠的射精潛伏期明顯長(zhǎng)于對(duì)照組(P<0.05),血清睪酮水平明顯低于對(duì)照組(P<0.05);帕羅西汀加1.0 mL洋蔥汁組、帕羅西汀加2.0 mL洋蔥汁組大鼠的騎跨潛伏期和帕羅西汀加2.0 mL洋蔥汁組大鼠的騎跨次數(shù)均明顯少于對(duì)照組和帕羅西汀組(P均<0.05);帕羅西汀加1.0 mL洋蔥汁組、帕羅西汀加2.0 mL洋蔥汁組大鼠的射精潛伏期均明顯低于帕羅西汀組(P均<0.05),血清睪酮水平均明顯高于帕羅西汀組(P均<0.05)。結(jié)論 新鮮洋蔥汁可以明顯增強(qiáng)雄性大鼠的性功能,并可以通過提高血清睪酮水平來緩解帕羅西汀引起的性功能障礙,增強(qiáng)性交配行為。

勃起功能障礙(Erectile dysfunction,ED)又稱為性無能,被定義為無法實(shí)現(xiàn)或保持陰莖勃起而滿足性生活需要的一種障礙。ED是一個(gè)世界性的問題,患病率較高,近年來中草藥用于ED的治療獲得了廣泛關(guān)注[1]。根據(jù)世界衛(wèi)生組織調(diào)查,在亞洲和非洲的一些國(guó)家,80%以上的人口依賴于傳統(tǒng)中草藥的替代醫(yī)學(xué)治療。Malviya等[2]研究證實(shí),洋蔥頭中提取的乙酸乙酯提取物可以恢復(fù)性功能障礙大鼠的正常性行為,然而目前其具體機(jī)制還不是很清楚。本研究觀察了不同劑量洋蔥汁對(duì)正常雄性大鼠和帕羅西汀所致性功能障礙雄性大鼠性功能和血清睪酮水平的影響,旨在探討洋蔥汁對(duì)大鼠性功能的影響機(jī)制,為后期臨床研究提供實(shí)驗(yàn)依據(jù)。
1.1 實(shí)驗(yàn)動(dòng)物 選取6月齡清潔級(jí)健康成年雄性SD大鼠80只,體質(zhì)量(250±20)g;雌性SD大鼠40只,體質(zhì)量(200±15)g。動(dòng)物質(zhì)量合格證號(hào):SCXK(鄂)2014-0012。大鼠均飼養(yǎng)于湖北醫(yī)藥學(xué)院動(dòng)物實(shí)驗(yàn)中心(SPF級(jí)),環(huán)境溫度控制在(21±1)℃,12 h的白天-黑夜光照周期循環(huán),給予標(biāo)準(zhǔn)飼料,自由飲食、飲水及活動(dòng),適應(yīng)性喂養(yǎng)2周。
1.2 動(dòng)物篩選 雌性SD大鼠在性交配實(shí)驗(yàn)前48 h內(nèi)注射苯甲酸雌二醇(上海通用藥業(yè)股份有限公司)50 mg促使發(fā)情,性交配實(shí)驗(yàn)前1 h內(nèi)注射黃體酮(浙江仙琚制藥股份有限公司)1 mg催情。對(duì)雄性大鼠進(jìn)行7次性行為交配試驗(yàn),選取在30 min內(nèi)完成射精性行為≥3次的雄性大鼠70只進(jìn)行實(shí)驗(yàn)。
1.3 藥物制備 新鮮洋蔥汁制備:新鮮洋蔥頭取自于山東省濰坊市濰北農(nóng)場(chǎng),去皮后用水果榨汁機(jī)(AJE-389.sona,中國(guó))提取制備;帕羅西汀懸浮液制備:帕羅西汀片購于中美天津史克制藥公司(國(guó)藥準(zhǔn)字H10950043,20 mg),采用1 mL蒸餾水按10 mg/kg濃度配置。
1.4 分組及給藥 將篩選出的70只雄性大鼠隨機(jī)分為7組,每組10只。每日16:00—17:00,對(duì)照組給予蒸餾水1 mL灌胃,0.5 mL洋蔥汁組、1.0 mL洋蔥汁組血清睪酮水平與2.0 mL洋蔥汁組分別給予相應(yīng)劑量的新鮮洋蔥汁灌胃,帕羅西汀組給予帕羅西汀懸浮液10 mg/kg灌胃,帕羅西汀加1.0 mL洋蔥汁組血清睪酮水平與帕羅西汀加2.0 mL洋蔥汁組分別給予帕羅西汀懸浮液10 mg/kg和相應(yīng)劑量的新鮮洋蔥汁灌胃,均1次/d,連續(xù)灌胃20 d。
1.5 觀察指標(biāo)
1.5.1 性行為參數(shù) 雄性大鼠的實(shí)驗(yàn)交配行為由2位毫不知情的實(shí)驗(yàn)觀察者記錄。于灌胃結(jié)束后的19:00—23:00,將雄鼠單獨(dú)放置在一個(gè)矩形的45 cm×40 cm×30 cm玻璃觀察室,微弱暗紅光照明,進(jìn)行5 min的適應(yīng)后,輕輕放入發(fā)情后的雌性大鼠,隨即攝錄下30 min內(nèi)大鼠的性交配行為表現(xiàn)。統(tǒng)計(jì)各組大鼠的性行為參數(shù),包括騎跨潛伏期(ML,即與雌鼠同籠至第1次騎跨所需時(shí)間)、插入潛伏期(IL,即與雌鼠同籠至第1次插入陰道所需時(shí)間)、騎跨次數(shù)(MF,即與雌鼠同籠從騎跨到射精前進(jìn)行的騎跨次數(shù))、插入次數(shù)(IF,即射精前插入雌性大鼠陰道次數(shù))、射精潛伏期(EL,即首次插入陰道到射精所需時(shí)間)、射精間隔時(shí)間(PEI,即從上次射精時(shí)間到下次插入時(shí)間)、交配有效率(CE,即IF/MF+IF)。
1.5.2 睪酮水平 上述各組大鼠在交配實(shí)驗(yàn)結(jié)束后24 h內(nèi)稱質(zhì)量,乙醚誘導(dǎo)麻醉后行心臟穿刺獲得血清,收集到離心管中,檢測(cè)血清睪酮水平。
2.1 各組雄性大鼠性行為參數(shù)比較 各組中大鼠30 min內(nèi)射精率均為100%。1.0 mL洋蔥汁組和2.0 mL洋蔥汁組大鼠的騎跨潛伏期、騎跨次數(shù)均明顯低于對(duì)照組和0.5 mL洋蔥汁組(P均<0.05),交配有效率均明顯高于對(duì)照組和0.5 mL洋蔥汁組(P均<0.05);1.0 mL洋蔥汁組和2.0 mL洋蔥汁組大鼠的插入潛伏期和1.0 mL洋蔥汁組大鼠的射精潛伏期均明顯低于對(duì)照組(P均<0.05);1.0 mL洋蔥汁組大鼠的插入潛伏期和射精潛伏期均明顯低于0.5 mL洋蔥汁組(P均<0.05);0.5 mL洋蔥汁組各性行為參數(shù)與對(duì)照組比較差異均無統(tǒng)計(jì)學(xué)意義(P均>0.05)。帕羅西汀組大鼠的射精潛伏期明顯長(zhǎng)于對(duì)照組(P<0.05);帕羅西汀加1.0 mL洋蔥汁組、帕羅西汀加2.0 mL洋蔥汁組大鼠的騎跨潛伏期和帕羅西汀加2.0 mL洋蔥汁組大鼠的騎跨次數(shù)均明顯少于對(duì)照組和帕羅西汀組(P均<0.05);帕羅西汀加1.0 mL洋蔥汁組、帕羅西汀加2.0 mL洋蔥汁組大鼠的射精潛伏期均明顯低于帕羅西汀組(P均<0.05);其余指標(biāo)各組間比較差異均無統(tǒng)計(jì)學(xué)意義(P均>0.05)。見表1。
2.2 各組大鼠血清睪酮水平比較 對(duì)照組、0.5 mL洋蔥汁組、1.0 mL洋蔥汁組、2.0 mL洋蔥汁組、帕羅西汀組、帕羅西汀加1.0 mL洋蔥汁組、帕羅西汀加2.0 mL洋蔥汁組血清睪酮水平分別為(5.34±0.42)ng/mL、(5.86±0.57)ng/mL 、(8.22±0.77)ng/mL 、(8.14±1.13)ng/mL 、(1.83±0.32)ng/mL 、(4.43±0.72)ng/mL 、(5.03±0.37)ng/mL 。1.0 mL洋蔥汁組、2.0 mL洋蔥汁組大鼠血清睪酮水平均明顯高于對(duì)照組和0.5 mL洋蔥汁組(P均<0.05),1.0 mL洋蔥汁組血清睪酮水平與2.0 mL洋蔥汁組比較差異無統(tǒng)計(jì)學(xué)意義(P均>0.05);帕羅西汀組大鼠血清睪酮水平明顯低于對(duì)照組(P<0.05),帕羅西汀加1.0 mL洋蔥汁組、帕羅西汀加2.0 mL洋蔥汁組血清睪酮水平均明顯高于帕羅西汀組(P均<0.05),帕羅西汀加1.0 mL洋蔥汁組血清睪酮水平與帕羅西汀加2.0 mL洋蔥汁組比較差異無統(tǒng)計(jì)學(xué)意義(P均>0.05)。

洋蔥是蔥屬百合科中最常見的一種球莖狀植物,并作為一種蔬菜在世界各地被廣泛種植。它常見于中東和印度及我國(guó)北方大部分地區(qū)人們的飲食中,由于其對(duì)人體健康具有多種益處而逐漸引起醫(yī)學(xué)界的關(guān)注。有研究證實(shí),洋蔥含有多種抗氧化物質(zhì),如硒、谷胱甘肽及維生素A、B、C等,這些物質(zhì)有助于保護(hù)細(xì)胞基因及其他分子免遭破壞和氧化[3]。Kumari等[4]報(bào)道從洋蔥中分離提出的S-甲基-L-半胱氨酸可明顯控制糖尿病大鼠的血糖水平。此外,洋蔥中還發(fā)現(xiàn)存在一些高濃度的微量元素礦物質(zhì)如銅、鋅,有助于體內(nèi)雄性睪丸激素的分泌。最近研究表明,新鮮的洋蔥汁0.5或1.0 g/d連續(xù)給藥20 d,可顯著提高雄性大鼠血清睪酮水平,提高大鼠的生精能力和性功能[5]。本研究結(jié)果顯示,1.0 mL洋蔥汁組和2.0 mL洋蔥汁組大鼠的騎跨潛伏期、騎跨次數(shù)均明顯低于對(duì)照組。騎跨潛伏期縮短說明大鼠的性渴望增強(qiáng),較早地表現(xiàn)出騎跨行為,而大鼠的騎跨次數(shù)減少直接反映了大鼠的性功能增強(qiáng),從而說明新鮮洋蔥汁可以改善雄性大鼠的性行為和增強(qiáng)性功能。
5-羥色胺受體是一種重要的神經(jīng)遞質(zhì),能夠產(chǎn)生一系列生理作用,幾乎影響到人體大腦活動(dòng)的各個(gè)方面,參與睡眠、痛覺及體溫等生理功能的調(diào)節(jié),中樞神經(jīng)系統(tǒng)5-HT含量增加,能使人產(chǎn)生一種積極的愉悅情緒,可明顯加速射精[6]。而帕羅西汀是一種選擇性5-羥色胺再攝取抑制劑,其通過增加細(xì)胞外的5-羥色胺水平而發(fā)揮作用,主要用途為抗抑郁,與其他抗抑郁類藥物一樣,會(huì)引起性功能障礙[7-8]。吳莉珍[9]報(bào)道,目前幾種新型的選擇性5-羥色胺再攝取抑制劑抗抑郁藥中,對(duì)抑郁患者性功能影響最大的為帕羅西汀,并且其發(fā)生障礙的程度與用藥劑量密切相關(guān)。 本實(shí)驗(yàn)結(jié)果顯示,帕羅西汀組大鼠的射精潛伏期明顯長(zhǎng)于對(duì)照組,與文獻(xiàn)[10]報(bào)道一致;帕羅西汀加1.0 mL洋蔥汁組、帕羅西汀加2.0 mL洋蔥汁組大鼠的騎跨潛伏期和帕羅西汀加2.0 mL洋蔥汁組大鼠的騎跨次數(shù)均明顯少于對(duì)照組和帕羅西汀組;帕羅西汀加1.0 mL洋蔥汁組、帕羅西汀加2.0 mL洋蔥汁組大鼠的射精潛伏期均明顯低于帕羅西汀組。提示新鮮洋蔥汁一定程度上可以緩解因帕羅西汀所致的延遲射精的現(xiàn)象,增強(qiáng)其性動(dòng)機(jī)。睪酮是主要的雄性激素,負(fù)責(zé)增強(qiáng)性行為和改善勃起功能。既往研究已經(jīng)表明,成人男性的血清睪酮水平輕微增加便可顯著增強(qiáng)男性性渴求與性欲望[11-12]。本研究結(jié)果顯示,1.0 mL洋蔥汁組、2.0 mL洋蔥汁組大鼠血清睪酮水平均明顯高于對(duì)照組和0.5 mL洋蔥汁組,1.0 mL洋蔥汁組、2.0 mL洋蔥汁組比較差異無統(tǒng)計(jì)學(xué)意義;帕羅西汀組大鼠血清睪酮水平明顯低于對(duì)照組,帕羅西汀加1.0 mL洋蔥汁組、帕羅西汀加2.0 mL洋蔥汁組血清睪酮水平均明顯高于帕羅西汀組,帕羅西汀加1.0 mL洋蔥汁組、帕羅西汀加2.0 mL洋蔥汁組比較差異無統(tǒng)計(jì)學(xué)意義。提示新鮮洋蔥汁可以增強(qiáng)體內(nèi)雄性激素的分泌,但血清睪酮水平并沒有隨著洋蔥汁劑量的增加而改變,其影響血清睪酮水平及刺激睪丸雄性激素的分泌是否有一個(gè)閾值劑量需要后期進(jìn)一步研究。
綜上所述,新鮮洋蔥汁可以增強(qiáng)雄性大鼠的性欲和性功能,并可以通過提高血清睪酮水平來緩解帕羅西汀引起的性功能障礙,增強(qiáng)性交配行為。后期將進(jìn)一步進(jìn)行臨床試驗(yàn),以證實(shí)新鮮洋蔥汁對(duì)人體的作用。

Study on the effect of fresh onion juice on copulatory behavior in male rats with paroxetine-induced sexual dysfunction and its mechanism

Objective It is to investigate the effects of onion juice on copulatory behavior and serum testosterone of sexually potent male rats and in male rats with paroxetine-induced sexual dysfunction, and provide experimental basis for clinical research in the later period. Methods 70 cases of male rats with normal sexual capacity were divided into seven groups, 10 cases in each group. The animals were treated with distilled water gavage in the control group, and with 0.5, 1.0 and 2.0 mL onion juice gavage in the corresponding dose onion juice groups, with 10 mg/kg paroxetine in the paroxetine group, with 10 mg/kg paroxetine plus 1.0, 2.0 mL onion juice in the corresponding dose groups, once a day, continuous treated for 20 days. At the end of the treatments, sexual behavior parameters and testosterone levels were measured and compared among all the groups. Results The incubation period and the times of riding in the 1.0, 2.0 mL onion juice group were significantly lower than those in the control group and the 0.5 mL onion juice group (allP<0.05), the efficiency of mating and serum testosterone levels were significantly higher than those in the control group and the 0.5 mL onion juice group (allP<0.05); the incubation period of insertion in the 1.0, 2.0 mL onion juice group and the incubation period of ejaculation in the 1.0 mL onion juice group were significantly lower than those in the control group (allP<0.05); the incubation period of insertion and ejaculation in the 1.0 mL onion juice group were significantly lower than those in the 0.5 mL onion juice group (allP<0.05). The incubation period of ejaculation in the paroxetine group was significantly longer than the control group (P<0.05), the level of serum testosterone was significantly lower than that in the control group (P<0.05). The incubation period of riding in the paroxetine plus 1.0 mL onion juice group and the times of riding in the paroxetine plus 2.0 mL onion juice group were significantly less than those in the control group and the paroxetine group (allP<0.05); the incubation period of ejaculation in the paroxetine plus 1.0, 2.0 mL onion juice group were significantly lower than those in the paroxetine group (allP<0.05), the level of serum testosterone was significantly higher than that in the paroxetine group (P<0.05). Conclusion The fresh onion juice can improve copulatory behavior of male rats, and in those with paroxetine-induced sexual dysfunction by increasing serum testosterone levels. Fresh onion juice can obviously enhance the sexual function of male rats, and it can alleviate the sexual dysfunction caused by paroxetine and enhance mating behavior by raising the level of serum testosterone.
