摘 要:研究胰島素樣生長因子-I(IGF-I)、肝細(xì)胞生長因子(HGF)、轉(zhuǎn)化生長因子β1(TGF-β1)、干擾素-γ(IFN-γ)對奶牛乳腺上皮細(xì)胞體外增殖的調(diào)節(jié)作用。應(yīng)用組織塊培養(yǎng)和差速消化法獲取原代奶牛乳腺上皮細(xì)胞,免疫熒光鑒定正確后MTT法評價不同濃度的4種細(xì)胞因子對奶牛乳腺上皮細(xì)胞體外增殖的影響。IGF-I、HGF分別在10~200 ng/mL,0.1~100 ng/mL濃度范圍內(nèi)對乳腺上皮細(xì)胞的增殖呈正相關(guān)。而TGF-β1(2.5~100 ng/mL)、IFN-γ(5~160 ng/mL)則劑量依賴性地抑制乳腺上皮細(xì)胞增殖。IGF-I、HGF均能促進(jìn)奶牛乳腺上皮細(xì)胞的體外增殖,該作用在一定濃度范圍內(nèi)有劑量依賴性;TGF-β1、IFN-γ對乳腺上皮細(xì)胞的增殖作用出現(xiàn)劑量抑制效應(yīng)。該研究為下一步深入探討細(xì)胞因子對奶牛泌乳機(jī)制的研究奠定基礎(chǔ)。

Effects of IGF-I、HGF、TGF-β1、IFN-γ on Proliferation in Bovine Mammary Epithelial Cells

Abstract:To investigate the regulation of IGF-I, HGF, TGF-β1 and IFN-γ in the growth of bovine mammary epithelial cells in vitro. The bovine mammary epithelial cells were isolated by explant culture technique and differential adhesion method and identified by immunochemistry.The effect of cytokines regulated the growth of bovine mammary epithelial cells in different concentrations was measured by MTT.When the concentrations of IGF-I and HGF were between 2.5~100 ng/mL and 5~160 ng/mL, the growth of bovine mammary epithelial cells increased. While When the concentrations of TGF-β1 and IFN-γwere between 2.5~100 ng/mL and 5~160 ng/mL, the growth of bovine mammary epithelial cells decreased in a dose-dependent manner.IGF-I and HGF could promote the bovine mammary epithelial cells grow in vitro in a dose-dependent manner.TGF-β1 and IFN-γcould inhibit the growth of bovine mammary epithelial cells in vitro.The information in this study may be useful to further study in the mechanism of effects of cytokine in lactation in dairy cow.
