酵母源生物飼料在畜牧業(yè)中的應(yīng)用較為廣泛,其產(chǎn)品多樣,通常劃分為飼料酵母、活性酵母、酵母培養(yǎng)物、酵母細(xì)胞壁多糖、酵母水解物、復(fù)合酵母以及酵母作載體,表達(dá)優(yōu)秀外源基因的“生物合成”產(chǎn)品。
1.2.1 干燥和包裝
活性干酵母實際上是處于“冬眠狀態(tài)”,并且其是活的生物體,遇到水和糖分后,就會“蘇醒”,繁殖生長。實際生產(chǎn)中,為了保持其活性,需要采用現(xiàn)代生物技術(shù)和先進(jìn)設(shè)備將生產(chǎn)的酵母細(xì)胞干燥成水分在5%以下的產(chǎn)品。經(jīng)過嚴(yán)格的干燥過程,利用真空或填充惰性氣體包裝的干酵母可以在1年內(nèi)保持其活性。如果未按要求生產(chǎn),活性干酵母的活性下降很快,品質(zhì)大大降低。
1.2.2 菌種
酵母種類有1 000多種,但并非都能發(fā)揮其特有的功能。Newbold等研究發(fā)現(xiàn),釀酒酵母菌株NCYC240,NCYC1026和商業(yè)制劑Yea-sac,能刺激瘤胃模擬裝置中總厭氧菌和纖維分解菌的生長,而NCYC694和NCYC1088則無此功能[1]。
1.2.3 日糧類型
在動物體內(nèi)酵母是作為活的生命體發(fā)揮作用的,動物腸道內(nèi)食糜是其發(fā)酵基質(zhì),所以不同的日糧組成會影響其發(fā)酵,由此產(chǎn)生的活性物質(zhì)也會有所差異。反芻動物的精料組成、纖維來源以及單胃動物日糧組成、抗生素的類型及數(shù)量都會影響活酵母的作用效果。即使同一產(chǎn)品的活酵母菌制劑在相同階段、同種動物上的應(yīng)用結(jié)果也不盡一致,可能主要受日糧組成的影響[2-6]。
活性酵母進(jìn)入瘤胃后會大量消耗瘤胃內(nèi)氧氣,降低氧濃度,從而提高厭氧菌數(shù)量,特別是能有效激活乳酸利用菌和纖維素分解菌,增加反芻動物的采食量,提高家畜對粗飼料的利用率,從而對生產(chǎn)性能起到改善作用[7-10]。Maloney等在玉米-豆粕型日糧中添加百福菌SC47飼喂斷奶仔豬,使仔豬日增重增加,改善飼料轉(zhuǎn)化率[2]。Heugten等在日糧中使用同種產(chǎn)品與高銅鋅及抗生素組對比,發(fā)現(xiàn)高銅鋅及抗生素組日糧同時添加酵母使仔豬采食量、平均日增重均提高[11]。但Hancock等和Hildabrand等向斷奶仔豬日糧中單獨添加酵母菌或與卡巴氧聯(lián)用未能顯著改善仔豬的生產(chǎn)性能[3,5]。Hildabrand等將酵母菌與硫酸新霉素聯(lián)用添加到飼料中顯著提高了仔豬生產(chǎn)性能[4]。雷宏俊等使用布拉啤酒酵母飼喂妊娠和哺乳母豬,降低了斷奶時母豬的體重?fù)p失,顯著降低了斷奶仔豬死亡率,提高了哺乳期母豬采食量[12]。Onifade等使用活性干酵母飼喂肉雞,提高了其養(yǎng)分消化率及平均增重、料肉比[13]。Thrune等給哺乳晚期的荷斯坦奶牛飼喂活性干釀酒酵母,能夠提高其瘤胃內(nèi)pH,從而降低亞急性瘤胃酸中毒的發(fā)生,以及瘤胃內(nèi)總VFA的值[14]。Seymour等在斷奶前后的犢牛料中加入面包酵母,可降低犢牛的發(fā)病率[15]。Adams等在荷斯坦奶牛料中使用活性酵母 Yea-Saccl026,結(jié)果發(fā)現(xiàn)對其產(chǎn)奶量產(chǎn)生積極影響[16]。Gtiger-Reverdin等在奶山羊飼料中加入啤酒酵母,可使低蛋白日糧組的奶山羊產(chǎn)奶量與對照組奶山羊的產(chǎn)奶量相當(dāng)[17]。Wohlt等在荷斯坦奶牛飼料中加入速酵源,顯著提高了哺乳初期奶牛干物質(zhì)進(jìn)食量、產(chǎn)奶量以及粗蛋白和酸性洗滌纖維的消化率[18]。Hill等在玉米-豆粕型日糧中加入活酵母飼喂荷斯坦、娟姍犢牛,結(jié)果表明,添加活酵母制劑4 g·d-1,63日齡娟姍犢牛末重更大[19]。Moallem等在小麥青貯飼糧中添加百福菌SC47,奶牛日產(chǎn)奶量提高4%,4%乳脂矯正奶提高6.1%。炎熱季節(jié)添加活酵母,一定程度上改善了瘤胃環(huán)境,提高了干物質(zhì)進(jìn)食量,并使生產(chǎn)性能和效率獲得提升[20]。Tomislav等在玉米-豆粕型日糧中添加百福菌飼喂哺乳晚期的克羅地亞雜交奶山羊,使奶羊第 23和27周產(chǎn)奶量顯著增加,整個試驗階段,酵母組奶山羊平均產(chǎn)奶量比對照組高(分別為630.28±92.34和 604.60±83.21),而且酵母組羊奶中乳脂比例顯著提高[21]。
酵母作為活的細(xì)胞前體,進(jìn)入胃腸道后繁殖和活力加強(qiáng),能有效抑制病原微生物的繁殖。原因可能是酵母菌在宿主體內(nèi)可以競爭性抑制病原菌對氧氣和營養(yǎng)物質(zhì)的攝入,從而降低有害菌的數(shù)量;酵母細(xì)胞壁外膜含有約甘露聚糖6.6%,其結(jié)構(gòu)類似腸道上皮細(xì)胞膜表面的甘露糖殘基,可競爭性的降低某些大腸桿菌和沙門氏菌對腸道的黏附[7,22];活酵母可以起到殺菌作用。王學(xué)東等在妊娠和哺乳母豬日糧中添加安琪活性干酵母,提高了哺乳母豬的采食量及斷奶仔豬成活率,并且改善了母豬胃內(nèi)菌群結(jié)構(gòu)[23]。潘寶海等使用釀酒酵母飼喂21日齡斷奶仔豬,結(jié)果表明,飼料中添加釀酒酵母可在一定程度上替代黃霉素等藥物添加劑。飼料中添加酵母制劑可改善21日齡斷奶仔豬消化道微生物區(qū)系,促進(jìn)有益菌的增殖,對大腸桿菌等有害菌的生長有抑制效果[24]。Line等給肉仔雞飼喂釀酒酵母后,以沙門氏菌和彎曲桿菌攻毒,并以轉(zhuǎn)運為應(yīng)激條件,結(jié)果顯示,應(yīng)激過程中沙門氏菌和彎曲桿菌在肉仔雞體內(nèi)定植的數(shù)量顯著降低[25]。Wohlt等在肉牛大麥日糧中添加酵母+微生態(tài)產(chǎn)品,結(jié)果顯示,酵母與細(xì)菌聯(lián)合使用在緩解瘤胃酸中毒和提高瘤胃發(fā)酵能力及營養(yǎng)物質(zhì)消化率方面無顯著作用[26]。
Buts等發(fā)現(xiàn),在人類志愿者和斷奶大鼠口服釀酒酵母后,小腸黏膜二糖酶的活性有所增強(qiáng)[27]。這對防止下痢、提高機(jī)體健康程度有積極意義。Buts等進(jìn)一步推斷,二糖酶活性受活酵母釋放的多胺(精胺和亞精胺)的調(diào)控[28]。
酵母細(xì)胞壁的葡聚糖和甘露聚糖在消化道內(nèi)會刺激網(wǎng)狀內(nèi)皮系統(tǒng),促進(jìn)機(jī)體產(chǎn)生巨噬細(xì)胞。在機(jī)體免疫系統(tǒng)中,巨噬細(xì)胞可以吞噬和清除體內(nèi)的異物、病原體和自身衰老細(xì)胞。Jurgens等將釀酒酵母應(yīng)用于玉米-豆粕型日糧飼喂仔豬,能夠提高其日增重和飼料報酬;飼喂妊娠母豬和哺乳母豬,提高了母豬乳中γ-球蛋白的含量[29]。
相對于其他無生命的酵母飼料或發(fā)酵培養(yǎng)物,活性干酵母是作為活菌制劑進(jìn)入動物體發(fā)揮作用的。除了本身具有一定的營養(yǎng)價值(酶、維生素、氨基酸和促生長因子)外,還會在機(jī)體代謝過程中產(chǎn)生多種有益活性物質(zhì),改善消化道微生物環(huán)境,促進(jìn)有益菌定植,提高動物的健康。活性酵母菌等益生菌不僅被廣泛用于飼料和飲水中,還被噴灑到畜舍內(nèi)。近年來發(fā)酵床養(yǎng)殖模式的成功,主要是靠微生物的分解與轉(zhuǎn)化能力,而活酵母菌一般作為制作發(fā)酵床的眾多菌種之一,與其他非真菌微生物一起協(xié)同作用,在養(yǎng)殖生態(tài)系統(tǒng)中完成物質(zhì)與能量的轉(zhuǎn)化。隨著人們對抗生素弊端認(rèn)識的不斷深入,活性酵母菌、乳酸菌和芽孢桿菌等在動物生產(chǎn)中利用最廣的益生微生物將越來越受人們的重視。
