2-芳基苯并呋喃類化合物在自然界中廣泛存在,同時具有多樣的生物活性,如抗腫瘤、抗病毒、抗真菌、抗氧化、腺苷A1拮抗、抗心肌缺血、抗炎、免疫抑制等[1-2]。2-芳基苯并呋喃類成分表現(xiàn)出巨大的藥用價值,極具開發(fā)潛力,長期以來備受關(guān)注[3]。Iteafuranal A(圖1)是從民族藥冬青葉鼠刺中分離鑒定的2-芳基苯并呋喃類化合物[4]。同時,進(jìn)一步的活性評價發(fā)現(xiàn)其表現(xiàn)出顯著的抗氧化和抗肝癌活性,可作為先導(dǎo)結(jié)構(gòu)進(jìn)一步開發(fā)[4-5]。
肼與羰基的縮合反應(yīng)即為腙化反應(yīng),是基于羰基開展結(jié)構(gòu)修飾的常用策略,可達(dá)到提升活性、降低毒性、發(fā)現(xiàn)新用途等目的[6-9]?;诖?,本研究以iteafuranal A為原料,合成了7個全新的苯腙衍生物,以期為后續(xù)先導(dǎo)結(jié)構(gòu)的發(fā)現(xiàn)奠定基礎(chǔ)。

Bruker Ascend 400 核磁共振儀(TMS為內(nèi)標(biāo),德國布魯克公司);Waters Xevo G2-S QTOF質(zhì)譜儀(美國Waters公司);ZF-7暗箱三用紫外分光儀(上海嘉鵬科技有限公司); N-1300D旋轉(zhuǎn)蒸發(fā)儀(東京理化器械株式會社);柱層析硅膠和 GF 254 薄層層析硅膠(青島海洋化工有限公司);所有試劑均為分析純,并經(jīng)蒸餾處理后使用。
參照現(xiàn)有方法進(jìn)行合成[10],具體步驟如下(圖2,以苯肼為例進(jìn)行描述):稱取iteafuranal A (1)30 mg(0.1 mmol),置于50 mL圓底燒瓶,以5 mL甲醇溶解。完畢,攪拌下加入苯肼32 mg(0.15 mmol)。接著,用膠頭滴管加入兩滴冰乙酸,60℃攪拌反應(yīng)。用TLC監(jiān)測反應(yīng)原料的反應(yīng)情況,直至反應(yīng)結(jié)束。通過硅膠柱層析,以二氯甲烷/甲醇(30∶1)洗脫得產(chǎn)物2a。2b-2g均參照此方法合成得到。

Iteafuranal A是從民族藥月月青分離得到的天然產(chǎn)物,具有3-醛基-2-芳基苯并呋喃的基本骨架,并表現(xiàn)出顯著的抗肝癌活性。其抗肝癌活性可能與靶向抑制RAS/ERK信號通路密切相關(guān),為新型靶向抗肝癌藥物的開發(fā)提供了新的先導(dǎo)結(jié)構(gòu)[5]。本研究基于其C-3位醛基官能團(tuán)展開結(jié)構(gòu)修飾研究,共合成了7個全新的苯腙衍生物,為進(jìn)一步的活性評價奠定堅實(shí)基礎(chǔ)。
