銀線草ChloranthusjaponicusSieb.又名燈籠花、分葉芹、假細(xì)辛,為金粟蘭科金粟蘭屬多年生草本植物。銀線草產(chǎn)于我國吉林、遼寧、河北、山東、山西、陜西、甘肅,亦分布于俄羅斯遠(yuǎn)東地區(qū)、朝鮮、日本。其生于林下陰濕處,根狀莖可提取芳香油,全株供藥用,具有祛濕散寒、活血止痛、散瘀解毒等功效,用于治療風(fēng)寒咳嗽、跌打損傷、癰腫瘡癤、婦女經(jīng)閉等[1]。研究表明,銀線草主要含有倍半萜、倍半萜多聚體和單萜類化學(xué)成分[2]?,F(xiàn)代藥理研究表明,銀線草提取物具有抗菌、抗人免疫缺陷病毒(HIV)和抗炎等生物活性[3]。本研究對(duì)銀線草化學(xué)成分進(jìn)行分離與鑒定,從銀線草75%乙醇提取物的乙酸乙酯部位共分離得到9個(gè)化合物,分別鑒定為金合歡素(1,acacetin)、千層紙素A(2,6-methoxybaicalein)、7E-3,4-二氧甲基苯并[2-苯基-胸苷]{3,7E-3,4-dioxymethylenzimtsaure-[2-phenyle-thylamid]}、落葉松脂素(4,lariciresinol)、9α-羥基紫菀內(nèi)酯(5,9α-hydroxyasterolide),銀線草醇B(6,shizukaol B)、金粟蘭素C(7,spicachlorantin C)、白術(shù)內(nèi)酯Ⅲ(8,atractylenolide Ⅲ)、白術(shù)內(nèi)酯Ⅱ(9,atractylenolide Ⅱ)。其中,化合物1~4為首次在金粟蘭屬中分離得到。
銀線草的全草采自陜西省安康市寧陜縣,經(jīng)華北理工大學(xué)藥學(xué)院莊鵬宇副教授鑒定為ChloranthusjaponicusSieb.,樣品留樣保存于華北理工大學(xué)藥學(xué)院天然藥物研究室(標(biāo)本號(hào):ID2016-1)。
硅藻土(煙臺(tái)化學(xué)研究所);75%乙醇、無水乙醇、石油醚、乙酸乙酯、二氯甲烷、甲醇、乙腈(天津天力化學(xué)試劑公司);純凈水(杭州娃哈哈集團(tuán)有限公司)。
JASCO-V650型紫外分析儀(北京金先鋒光電科技有限公司);1260型分析型高效液相色譜儀(美國Agilent公司);6AT型半自動(dòng)制備型高效液相色譜儀(日本島津公司);Avance ⅡHD 600型核磁共振儀(德國Bruker公司);柱色譜硅膠400~500目、薄層色譜硅膠H(青島海洋化工有限公司)。
銀線草全草(4 kg)干燥粉碎,用75%乙醇水回流提取(8倍量溶劑,回流4 h,2次),40 ℃減壓干燥得到48 g浸膏。浸膏用甲醇復(fù)溶吸附于等量的硅藻土上并完全干燥,依次用石油醚、乙酸乙酯進(jìn)行洗脫(4次),分別得到石油醚洗脫部分720 g和乙酸乙酯洗脫部分700 g。取乙酸乙酯洗脫部分進(jìn)行硅膠柱色譜分離,洗脫劑為二氯甲烷-甲醇,梯度洗脫(100∶0→0∶100),根據(jù)薄層色譜檢測結(jié)果進(jìn)行相似成分合并,得到Y(jié)EA~YEF 5個(gè)組分。YEA經(jīng)過凝膠柱色譜純化,甲醇-水(30∶70)洗脫,得到化合物1(12.5 mg)和2(6.1 mg);YEB經(jīng)過硅膠柱色譜(石油醚-乙酸乙酯,10∶1)洗脫,得到化合物3(5.2 mg)和4(7.6 mg);YEC經(jīng)過硅膠柱色譜(石油醚-乙酸乙酯,5∶1)洗脫,得到化合物5(20.6 mg)和6(40.8 mg);YED經(jīng)過凝膠柱色譜(二氯甲烷-石油醚-甲醇,4∶4∶1)洗脫,得到化合物7(25.6 mg)?;衔?母液經(jīng)半自動(dòng)制備液相純化(乙腈-水,45∶55)得到化合物8(5.2 mg)和9(2.3 mg)。

本研究在銀線草75%乙醇提取物的乙酸乙酯溶性部位中分離得到9個(gè)化合物,其中化合物1~4為首次在金粟蘭屬中分離得到,豐富了金粟蘭屬的化學(xué)成分,為其藥理活性研究提供參考。
