【摘要】 目的 探討地塞米松對小鼠血壓和心率的影響,尋找制備動物高血壓模型的藥物。
方法 取KM小鼠40只隨機分為正常對照組(給予0.9% NaCl注射液)和地塞米松低、中、高濃度組(分別給予1.25 mg/kg、2.5 mg/kg、5.0 mg/kg地塞米松注射液),肌肉注射每天一次(用量按小鼠體重10 mL/kg計算),連續(xù)14天。各組小鼠在末次用藥后20 min測定收縮壓、舒張壓和心率。
結(jié)果 正常對照組和地塞米松低濃度組的實驗后收縮壓、舒張壓、心率與實驗前比較差異均無統(tǒng)計學(xué)意義(P>0.05)。地塞米松中濃度組和高濃度組實驗后的收縮壓、舒張壓均明顯高于實驗前和正常對照組(P<0.05或0.01),地塞米松高濃度組實驗后的收縮壓、舒張壓均明顯高于地塞米松低濃度組、中濃度組(P<0.01)。正常對照組和地塞米松低濃度組、中濃度組的實驗前后心率兩兩比較差異無統(tǒng)計學(xué)意義(P>0.05)。地塞米松高濃度組的實驗后心率與正常對照組和地塞米松低濃度組、中濃度組比較差異無統(tǒng)計學(xué)意義(P>0.05),但明顯高于其實驗前(P<0.05)。
結(jié)論 地塞米松可加強心血管興奮引起升高血壓的作用,此作用存在量效關(guān)系;地塞米松用量達一定高濃度可加快心率。

【Abstract】 Objective To explore the effect of dexamethasone on blood pressure and heart rate in mice,and to search for drugs for the preparation of animal hypertension model.
Methods 40 KM mice were randomly divided into normal control group(given 0.9% NaCl injection) and dexamethasone low concentration group,dexamethasone medium concentration group and dexamethasone high concentration group(given 1.25 mg/kg,2.5 mg/kg and 5.0 mg/kg dexamethasone injection,respectively).Intramuscular injection was performed once a day(dosage was calculated according to 10 mL/kg of mouse body weight) for 14 consecutive days.And then,systolic blood pressure,diastolic blood pressure and heart rate were measured 20 min after the last administration in each group.
Results There was no statistically significant difference in systolic blood pressure(SBP),diastolic blood pressure(DBP) and heart rate(HR) between the normal control group and dexamethasone low concentration group before and after experiment(P>0.05).After experiment,the systolic and diastolic blood pressure of dexamethasone in the middle and high concentration groups were all significantly higher than those before experiment and those in the normal control group(P<0.05 or 0.01),and those in the dexamethasone high concentration group were significantly higher than those in the dexamethasone low concentration group and dexamethasone medium concentration group(P<0.01).There was no statistically significant difference in heart rates between the normal control group,the dexamethasone low concentration group and the dexamethasone medium concentration group before and after experiment(P>0.05).After experiment,there was no statistically significant difference in heart rate between the dexamethasone high concentration group, the normal control group,the dexamethasone low concentration group and the middle concentration group(P>0.05),but they were all higher than those before experiment(P<0.05).
本研究結(jié)果警示,使用地塞米松治療的患者應(yīng)加強對血壓的監(jiān)測,高血壓、冠心病、心腦血管意外的患者慎用。
